RESEARCH SUMMARY
Evidence of pathogenic effects has been found for antineutrophil cytoplasmic autoantibodies (ANCA), most notably those with antimyeloperoxidase or antiproteinase-3 specificity, in small-vessel vasculitides (Wegener's granulomatosis, Churg-Strauss syndrome, and microscopic polyangiitis). The disease process is thought to involve complex interactions between genetic and environmental factors leading to a humoral autoimmune response against neutrophilic components. My work explores potential pathomechanistic explanations of ANCA-associated diseases. Specifically, what is the origin of autoantibodies and how do environmental exposures contribute; do the autoantibody/granulocyte interactions cause pathogenic changes in granulocyte functions, including changes in gene expression profiles; and what factors are primary in the genesis of resultant endothelial lesions of the vascular endothelium?
